Description

QIAGEN offers LNA-enhanced detection probes designed for ultra-sensitive and specific in situ hybridization (ISH) of mRNA and small RNAs. The proprietary LNA technology modifies bases at selected positions of mRNA, lncRNA, and circRNA probe sequences without altering the sequence itself, enabling Tm equalization across probes while maintaining single-base specificity and sensitivity. Custom probes for any mRNA or lncRNA target can be configured through advanced online design tools, allowing researchers to tailor each probe to their specific application.

These probes are suited to detecting short RNAs and discriminating between highly similar RNAs. Their high affinity for target sequences permits the use of stringent hybridization and washing conditions even when short probe lengths are required. Common labeling options include digoxigenin, biotin, amino, and fluorescent dyes, supporting both single-end and double-end labeling strategies.

Among the fluorescent labels available are FAM (equivalent to FITC) at 495/520 nm (green), TYE563 (equivalent to Cy3) at 549/563 nm (yellow), and TYE665 (equivalent to Cy5) at 645/665 nm (red). Double-end labeling produces greater signal amplification than single-end labeling, though at higher cost. The technology supports challenging sample types, with related buffer sets recommended for FFPE workflows.
