ArciTect HiFi DNA Pol Kit

Engineered for robust, high-fidelity DNA amplification, the ArciTect™ High-Fidelity DNA Polymerase Kit from STEMCELL Technologies delivers a 50- to 60-fold lower error rate than standard Taq polymerase as determined by the lacI mutagenesis assay. The enzyme is a fusion protein featuring a double-stranded DNA-binding domain and a Pyrococcus-like proofreading polymerase domain, and it generates blunt-end products. It is suitable for sequencing, cloning and subcloning, synthetic biology, SNP analysis, and for the detection of genome editing when used with the ArciTect™ T7 Endonuclease I Kit.

All kit components are animal component-free. The kit contains the ArciTect™ High-Fidelity DNA Polymerase, a 10 mM dNTP Mix containing dATP, dCTP, dGTP, and dTTP sodium salts, and two reaction buffers: ArciTect™ High-Fidelity Buffer for standard high-fidelity amplifications and ArciTect™ High GC Content Buffer for GC-rich or difficult-to-amplify templates. Reactions using the High GC Content Buffer provide increased sensitivity with a slightly higher error rate. The polymerase is expressed in E. coli, has a molecular weight of 97.7 kDa, and is supplied at 2 U/μL. It exhibits 3' to 5' proofreading exonuclease activity but lacks 5' to 3' nick-translation activity.

The polymerase offers an extension rate of 67 nucleotides per second and successfully amplifies long targets, including up to 5 kb of human genomic DNA and 8 kb of lambda DNA. The product can be used in conventional PCR but not in real-time or digital PCR. All components should be stored at -20°C and remain stable until the expiry date on the label. The kit is for research use only.
