Description

From QIAGEN, this Antisense LNA GapmeR is supplied as the in vitro screening grade variant with standard desalting purification, making it suitable for testing experiments across various designs. Antisense LNA GapmeRs are antisense oligonucleotides that efficiently suppress the function of mRNA and lncRNA. They are generally 14-16 nucleotides in length and fully phosphorothioated. The construct spans 16 nt, with LNA-modified ends flanking an internal DNA region, and recognizes and binds target mRNA and lncRNA. RNase H is recruited to the central DNA region to mediate degradation of the target RNA. The use of LNA technology provides high-affinity binding to the target site, helping to reduce off-target effects. Because the oligonucleotide is a nuclease-resistant nucleic acid analog that enters cells via cell membrane surface receptor-mediated uptake, it acts on both nuclear and cytoplasmic RNA — notably enabling knockdown of nuclear lncRNAs where conventional siRNA is not effective. The same chemistry supports use in both in vitro cell experiments and in vivo animal studies, with tissue-dependent half-lives in animals reaching up to 4 months at the longest.
