For sequence-specific mRNA knockdown in animal models, QIAGEN supplies the Antisense LNA GapmeR in vivo LS as a custom-designed antisense oligonucleotide. The construct pairs locked nucleic acid (LNA) residues at both ends with a central DNA gap, recruiting RNase H to cleave the target transcript. The LS formulation is engineered for systemic delivery and intracellular uptake, supporting intraperitoneal, subcutaneous, or intravenous dosing in rodents.

Unlike siRNA, GapmeRs do not depend on the RISC machinery, making them a reliable antisense alternative for target validation in vivo, preclinical evaluation of candidate genes, and functional studies in liver, tumor, and other tissues. Each order is delivered in a single-use vial containing 5 mg of purified material, typically adequate for several dosing cohorts depending on the route of administration and dose required per animal.

Handling requires reconstitution in nuclease-free water or PBS, followed by storage as frozen aliquots at -20 °C to protect against freeze-thaw degradation and preserve activity. The dried oligonucleotide is stable for extended periods when kept at the recommended storage temperature and shielded from light. Researchers submit the target accession to the QIAGEN design pipeline, which returns a fully optimized GapmeR ready for research use applications in vivo.
