Description

A QIAGEN negative control reagent designed for RNAi experiments, this siRNA has been validated using Affymetrix GeneChip arrays and cell-based assays. The reagent is supplied in tube format with chemical modification and is intended for use in human, mouse, and rat systems.

Genomewide profiling performed in MCF-7, K562, and primary HUVEC cells transfected with HiPerFect Transfection Reagent showed that this siRNA produced the lowest number of nonspecifically regulated genes compared with other negative control siRNAs tested, including nonsilencing, scrambled, and reporter-targeting sequences. Cell-based assays confirmed normal nuclear size, unchanged proliferation rates, unchanged DNA synthesis as measured by BrdU incorporation in HCT-116 cells, no increase in cytotoxicity, and a normal cell-cycle distribution relative to untransfected cells.

RISC-incorporation analysis in HeLa and MCF-7 cells confirmed that the siRNA enters RISC, validating it as a control that undergoes the same biological process as gene-specific siRNAs. In the supporting studies, regulated genes were defined as those showing at least a 1.5-fold change in expression versus untransfected cells. The reagent can be combined with other controls such as AllStars Cell Death Control siRNA, GAPDH, Beta-Actin, MAPK1, Lamin A/C, and AKT1 siRNAs on FlexiPlate siRNA plates.
