Description

A negative control siRNA from QIAGEN, intended for use in RNAi experiments as a baseline for evaluating gene-specific knockdown. According to the manufacturer's specifications, the reagent has been predesigned and validated using Affymetrix GeneChip Array analyses and cell-based assays, is supplied in tube format, and is suitable for human, mouse, and rat species. The product carries a modification.

Validation data demonstrate minimal nonspecific effects across multiple assay types. In genome-wide expression profiling conducted in MCF-7, K562, and primary HUVEC cells using HiPerFect Transfection Reagent, this control consistently produced the lowest number of nonspecifically regulated genes among negative control siRNAs tested. Cell-based assays showed that transfection did not change nuclear size, cell proliferation, DNA synthesis rates, cytotoxic effects, or cell-cycle distribution relative to untransfected cells. RISC-incorporation analysis in HeLa and MCF-7 cells confirmed that the siRNA is incorporated into RISC, indicating that it undergoes the same biological process as gene-specific siRNAs. In the profiling experiments, regulated genes were defined as those showing at least a 1.5-fold change in expression compared with untransfected cells.
