Designed for negative control RNAi experiments, this predesigned siRNA from QIAGEN has been validated by Affymetrix GeneChip Array and cell-based assays. It is suitable for human, mouse, and rat species, supplied in tube format, and includes modifications.

Validation studies confirmed minimal nonspecific regulation of gene expression. In genomewide analysis performed after transfection into MCF-7, K562, and primary HUVEC cells using HiPerFect Transfection Reagent, the control siRNA consistently produced the lowest number of nonspecifically regulated genes compared with other negative control siRNAs, including nonsilencing, scrambled, and reporter-gene–targeting sequences. Cell-based assays further demonstrated normal phenotypes: live-cell nuclei staining showed nuclear size remained comparable to untransfected cells; cell counts indicated unchanged proliferation rates; Bromodeoxyuridine (BrdU) nucleotide incorporation measurements showed unchanged DNA synthesis rates; live-cell dye exclusion showed no increase in cytotoxic effects; and DNA staining demonstrated normal cell-cycle distribution across G1/G0, S, and G2 phases.

RISC-incorporation analysis performed in HeLa and MCF-7 cells using reporter construct transfection confirmed that the siRNA enters RISC (RNA-Induced Silencing Complex), enabling it to serve as a reliable benchmark that undergoes the same biological processing as gene-specific siRNAs. Across the validation studies, regulated genes were defined as those showing at least a 1.5-fold change in expression compared with untransfected cells.
