Library preparation for next-generation sequencing relies on efficient adapter ligation, and QIAGEN supplies this 2.5x concentrated reagent as a ready-to-use buffer for that critical enzymatic step. The formulation delivers the ionic strength, pH buffering, and ATP cofactor conditions required for robust covalent joining of 5'-phosphorylated DNA ends to sequencing adapter oligonucleotides.

Added directly to ligation reactions containing fragmented DNA, sequencing adapters, and a DNA ligase, the 2.5x buffer simplifies reaction setup by eliminating the need to prepare fresh buffer from individual components. It is compatible with standard Y-shaped and forked Illumina-compatible adapters as well as Ion Torrent-compatible adapters, and supports both PCR-free and amplification-included library construction workflows including whole-genome sequencing, targeted resequencing, amplicon sequencing, and methylation sequencing.

  • Supplied as a 2.5x concentrated buffer for direct use in ligation reactions
  • Storage temperature: -30°C to -15°C in a frost-free freezer
  • Shelf life: 12 months from manufacture when handled per instructions and protected from repeated freeze-thaw cycles
  • Recommended reaction incubation: 30 minutes at room temperature with ligase
  • Format: single-use tube to minimize contamination risk and ensure reproducible performance

Researchers running enzymatic fragmentation followed by direct ligation will find the buffer's salt and ATP concentrations balanced to maintain ligase activity throughout the incubation window. The reagent integrates into QIAGEN's broader NGS consumables portfolio and can be combined with fragmentation, end-repair, and cleanup reagents for a complete library preparation workflow.
