Plasmid Buffer Set

A comprehensive collection of buffers and RNase A from QIAGEN supports plasmid DNA purification workflows when used alongside the QIAGEN Plasmid Mini, Midi and Maxi Kits.

The manufacturer Quick-Start Protocol states the buffer set can be stored at room temperature (15–25°C) for up to 2 years if not otherwise stated on the label. Before starting, RNase A solution is added to Buffer P1, mixed, and stored at 2–8°C. Optionally, LyseBlue reagent is added to Buffer P1 at a ratio of 1:1000. Buffer P3 is prechilled to 4°C, Buffer P2 is checked for SDS precipitation, and isopropanol and 70% ethanol are required as additional reagents.

The purification workflow involves harvesting bacterial culture by centrifugation at 6000 x g for 15 min at 4°C, resuspending the pellet in Buffer P1, lysing with Buffer P2 at room temperature (15–25°C) for 5 min, and neutralizing with prechilled Buffer P3. After centrifugation, the cleared lysate is applied to a QIAGEN-tip pre-equilibrated with Buffer QBT. After washing with Buffer QC, DNA is eluted with Buffer QF, precipitated with 0.7 volumes of room-temperature isopropanol, washed with 70% ethanol, and redissolved in a suitable buffer such as TE buffer (pH 8.0) or 10 mM Tris·Cl (pH 8.5).
