Solution CD3 (175 ml)

A high-concentration salt solution from QIAGEN, Solution CD3 is used as a reagent in the DNeasy PowerSoil Pro Kit workflow for isolating genomic DNA from environmental soil samples. According to the kit handbook, the solution contains chaotropic salts, which are added after the Inhibitor Removal Technology step to adjust the salt concentration of the DNA solution. Under these high-salt conditions, DNA binds tightly to the silica membrane in the MB Spin Column during purification. The standard protocol calls for adding 600 µl of Solution CD3 to the supernatant, followed by vortexing for 5 seconds.

If Solution CD3 precipitates, it can be re-dissolved by heating at 60°C. The solution contains chaotropic salts and should not be combined directly with bleach, as this can form highly reactive compounds. Spills should be cleaned first with a suitable laboratory detergent and water, and then with 1% (v/v) sodium hypochlorite if potentially infectious agents are present.
