The Antibodies.com recombinant Lambda Protein Phosphatase is a 25 kDa Mn2+-dependent serine/threonine/tyrosine phosphatase encoded by bacteriophage lambda, supplied as an active liquid enzyme in 5 x 20 kU vials. One unit hydrolyzes 1 nmol of p-nitrophenyl phosphate per minute at 30 °C in the supplied reaction buffer, providing reproducible removal of phosphate groups from phospho-Ser, phospho-Thr, and phospho-Tyr residues without the substrate bias commonly observed with calf intestinal alkaline phosphatase. Researchers routinely deploy this phosphatase to strip phosphate modifications from target proteins ahead of SDS-PAGE mobility-shift analysis, to back-phosphorylate kinase substrates in functional studies, and to validate phospho-specific antibody epitopes by abolishing immunoreactivity against the modified residue. The recombinant enzyme is reconstituted and diluted immediately before use into buffer supplemented with 1 mM MnCl2, which is required for catalytic activity. Store the vial frozen at -20 °C in single-use aliquots to limit freeze-thaw loss of activity, where it retains full potency for at least 12 months under proper handling.
