The recombinant Ribonuclease H from Antibodies.com is an E. coli-derived endoribonuclease that specifically hydrolyzes the RNA strand of RNA:DNA hybrids, generating 5'-phosphate and 3'-hydroxyl termini without cleaving the DNA component. Expressed from the native rnhA gene, the enzyme is supplied as a 17.5 kDa monomer in liquid form and ships as a pack of 5 vials totaling 5,000 U of activity.

The enzyme requires a divalent cation such as Mg2+ or Mn2+ and functions optimally near 37 °C, supporting workflows such as second-strand cDNA removal during first-strand cDNA synthesis, R-loop mapping in transcription studies, cleavage of RNA primers during nucleic acid processing, and in vitro characterization of RNA:DNA hybrid substrates. Store the enzyme frozen at -20 °C and avoid excessive freeze-thaw cycles to preserve hydrolytic activity across downstream functional studies.

Each lot is verified by SDS-PAGE for purity and by a nuclease activity assay against a defined RNA:DNA hybrid substrate, ensuring reproducible unit activity between batches for molecular biology and enzymology research.
