A core consumable in the PyroMark Q24 pyrosequencing workflow from QIAGEN, this 250 ml bottle of PyroMark Annealing Buffer provides the optimized ionic environment required for the primer hybridization step. After biotinylated PCR products are converted to single-stranded DNA on the PyroMark Q24 Vacuum Workstation using PyroMark Streptavidin Sepharose beads, the buffer is dispensed into each well of the PyroMark Q24 Plate so the sequencing primer can anneal to the purified template. The reagent supports mutation analysis, methylation quantification, and genotyping assays, and the 250 ml format is sufficient for processing multiple 24-well plates. Each well receives a small working volume of buffer, supporting consistent hybridization before nucleotides, enzymes, and substrates are dispensed by the PyroMark Q24 cartridge for the run. Positioned between Vacuum Workstation template isolation and the final PyroMark Q24 instrument read, the buffer ensures that the primer is correctly annealed to the biotinylated template strand for downstream in vitro sequencing analysis.
