The p24 polypeptide is the 24 kDa capsid (CA) structural protein encoded by the HIV-1 gag gene; it self-assembles into the conical core that packages the viral RNA genome inside infectious virions. Because p24 is one of the earliest and most abundant viral proteins produced during active replication, the recombinant form is widely used to standardize ELISA plate coatings for serological screening and as a positive control band in Western blot confirmation of anti-Gag antibody responses. It is also used in vaccine research to map B-cell and T-cell epitopes within the conserved capsid region, in antibody-pair screening for p24-specific capture and detection reagents, and in assay development aimed at detecting acute infection during the pre-seroconversion window.
