A chaotropic lysis reagent supplied by QIAGEN in a 750 ml bottle, Buffer FG2 is the second solution used in the FlexiGene DNA purification workflow for extracting high-molecular-weight genomic DNA from whole blood, buffy coat, bone marrow, and cultured mammalian cells. After red blood cells are removed with Buffer FG1, the leukocyte pellet is resuspended in Buffer FG2, whose combination of guanidine hydrochloride, urea, and a nonionic detergent rapidly lyses cell membranes, denatures histones, and dissociates DNA-protein complexes to liberate intact genomic DNA. The lysate is then mixed with Buffer FG3 for protein precipitation, followed by isopropanol-driven DNA precipitation and an ethanol wash. The resulting DNA is typically several hundred kilobases in size and is suitable for downstream applications such as PCR, restriction enzyme digestion, Southern blotting, long-range amplification, SNP genotyping, and archival storage.

Store tightly capped at room temperature (15-25 degrees C); the reagent remains stable under these conditions for the duration of its shelf life and does not require refrigeration. Always handle with gloves and eye protection, since guanidine salts are irritants and should be discarded through an appropriate laboratory waste stream rather than combined with bleach in a closed vessel. The 750 ml bottle format supports processing of roughly 200 standard blood draws (3-10 ml each) at the buffer volumes specified in the FlexiGene handbook.
