Buffer EL

An erythrocyte lysis buffer from QIAGEN, Buffer EL is formulated to disrupt red blood cells in whole blood samples prior to white blood cell isolation and downstream genomic DNA extraction. Used as part of the Puregene Blood Kit workflow, the ammonium chloride-based solution rapidly lyses erythrocytes while preserving intact leukocytes for subsequent nucleic acid purification. Standard protocol application involves adding the reagent to anticoagulated whole blood, brief incubation, and centrifugation to collect the white blood cell pellet, which then proceeds through cell lysis, protein precipitation, and DNA hydration steps. The reagent is supplied in a ready-to-use liquid bottle format suited for routine laboratory workflows and high-throughput processing. Storage at room temperature maintains long-term reagent stability, and single-use aliquots for each blood sample minimize cross-contamination risk in research use only applications such as genotyping, DNA sequencing, and PCR-based studies.
