Buffer EL (2 x 250 ml)

This QIAGEN erythrocyte lysis buffer is intended for the selective lysis of erythrocytes from human blood and bone marrow aspirates, allowing subsequent recovery of leukocytes. The procedure is performed manually, with the buffer added to the sample and incubation carried out on ice.

Blood or bone marrow aspirates contain up to 1000 times more erythrocytes than leukocytes, so their removal simplifies downstream steps such as RNA isolation. Erythrocytes are more susceptible than leukocytes to hypotonic shock and burst rapidly in the hypotonic environment, while leukocytes remain stable and can be recovered intact by centrifugation. The product is intended for laboratory use by professional users such as technicians and physicians trained in molecular biological techniques, and may only be used in conjunction with QIAGEN kits that reference it in their respective instructions for use.

The product is an in vitro diagnostic medical device that fulfils the requirements of European Regulation (EU) 2017/746 (IVDR). It contains no active ingredients; lysis is achieved by a physical principle (osmosis) providing a hypotonic environment. It is shipped at ambient conditions and must be stored upright at room temperature (15–25°C); once opened, reagents can be kept in their original packaging at 15–25°C for up to 3 months, but not beyond the printed expiration date. Each lot is tested against predetermined specifications under QIAGEN's ISO-certified Quality Management System. The buffer is not classified as hazardous. Materials required but not provided include a laboratory centrifuge with a swinging bucket rotor capable of ≥400 x g at 4°C, 50 mL centrifuge tubes, and disposable gloves.
