MethoCult H4434 Classic, 24x3mL/tube

Methylcellulose-based colony-forming unit (CFU) assays remain the standard readout for human hematopoietic stem and progenitor cell biology. The H4434 Classic formulation from STEMCELL Technologies delivers a complete, ready-to-plate medium in 3 mL tubes (24 tubes per case), with each tube already containing the recombinant human cytokine cocktail needed to drive erythroid, myeloid, and multilineage colony growth from mononuclear cell sources.

Each 3 mL aliquot supplies SCF, FLT3 ligand, IL-3, IL-6, G-CSF, GM-CSF, and erythropoietin within a viscous methylcellulose matrix, supporting simultaneous scoring of BFU-E, CFU-GM, and CFU-GEMM colonies. Typical workflow: 1-5 x 10⁵ mononuclear cells per 35 mm non-tissue-culture-treated dish in approximately 1.1 mL of medium, incubated at 37 °C, 5% CO₂, and ≥95% humidity for 14 days, then counted under an inverted microscope.

Applications include characterization of mobilized peripheral blood or cord blood grafts, clonogenic recovery studies after cryopreservation, off-target cytotoxicity screening of candidate compounds, and lineage-commitment investigations in disease-model or gene-editing workflows. Tubes must be stored frozen at -20 °C and thawed at 4 °C immediately before plating; refreezing thawed aliquots is not recommended. For research use only.
