Methylcellulose-based hematopoietic colony assay medium from STEMCELL Technologies enables enumeration of erythroid and myeloid progenitor cells in a ready-to-use format. This in vitro research formulation supports growth and quantification of burst-forming unit erythroid (BFU-E), colony-forming unit granulocyte/macrophage (CFU-GM), and multi-lineage CFU-GEMM colonies from human bone marrow, cord blood, or mobilized peripheral blood mononuclear cells in a single tube aliquot. Researchers plate cells directly into the viscous medium without further mixing or supplementation, and scoring at day 14 under an inverted microscope yields a colony count per well. The methylcellulose matrix holds cells in a semi-solid layer that mimics the bone marrow microenvironment. Each vial is intended for single use, shipped frozen on dry ice, and stored at -20 °C protected from light; thaw only what is needed to limit freeze-thaw cycles. The Express format eliminates the powder rehydration step that older MethoCult formulations require, reducing hands-on time and inter-lot variability. Common downstream uses include compound screening for hematotoxicity, characterization of cord-blood units, and studies of cytokine-driven differentiation in CD34+ enriched populations. Cell culture applications in academic and translational hematology labs rely on this standardized medium for reproducible colony output.
