The assay uses an indirect ELISA format in which TOB1 (phospho Ser164) is captured by anti-TOB1 (phospho Ser164) antibodies and detected with HRP-conjugated secondary antibodies, producing a colorimetric signal upon substrate addition. As a qualitative cell-based ELISA, the kit provides three normalization approaches: an anti-GAPDH antibody as an internal positive control, Crystal Violet whole-cell staining to determine cell density, and an anti-TOB1 antibody for normalizing the phosphorylated target absorbance against total TOB1 values. Detection is colorimetric on a tissue culture treated microplate platform, with a total assay time of 4 hours 30 minutes.
