Description
A cell-based, qualitative immunoassay from Antibodies.com is designed for the indirect in vitro quantitative determination of SHC (phospho Tyr427) levels in adherent and suspension cells. SHC is also known by the synonyms SH2 domain protein C1, SHC-transforming protein 1, SHC-transforming protein 3, SHC-transforming protein A, SHC1, SHCA and Src homology 2 domain-containing-transforming protein C1.
The assay works by capturing Shc (phospho Tyr427) with anti-Shc (phospho Tyr427) antibodies, which are in turn detected by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The readout is performed on a tissue culture treated microplate. Reactivity is reported for human (Tyr427), mouse (Tyr423) and rat (Tyr313) samples, with no reported cross-reactivity, and the assay is suitable for a range of more than 5000 cells. Three normalization approaches are included: an anti-GAPDH antibody as an internal positive control, Crystal Violet whole-cell staining to adjust absorbance values to cell density, and an anti-Shc antibody that allows the phosphorylated target signal to be normalized against the non-phosphorylated target.
The complete protocol takes approximately 4 hours and 30 minutes. The kit is stored at +4 °C and is reported to remain stable for 3 to 6 months.
