This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of rat Wolfram Syndrome Protein 1 (WFS1) in tissue homogenates or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for WFS1. Standards and samples are added to the wells, followed by a Biotinylated Anti-WFS1 Antibody and an HRP-Avidin conjugate. A TMB substrate is added and the resulting colour is read at 450 nm, with WFS1 concentration calculated by reference to the standard curve. The assay has a measuring range of 0.312–20 ng/ml and a sensitivity of <0.118 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Detection is performed on a colourmetric microplate reader.

Recovery values reported for the kit are 80–102% (average 91%, n=5) in serum, 81–100% (average 90%, n=5) in EDTA plasma, and 80–89% (average 84%, n=5) in heparin plasma. Linearity across dilutions from 1:2 to 1:16 has been assessed, with serum samples (n=5) showing recovery ranges of 87–91%, 87–107%, 74–101% and 92–97% at the four dilutions respectively; EDTA plasma (n=5) showing 90–105%, 84–101%, 90–101% and 79–108%; and heparin plasma (n=5) showing 84–95%, 92–105%, 82–105% and 89–91%. The kit includes a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
