Rat Wingless Type MMTV Integration Site Family, Member 3 ELISA Kit

A sandwich (quantitative) Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of rat Wnt3 in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated microplate bearing an antibody specific for Wnt3; bound analyte is detected with a Biotinylated Anti-Wnt3 Antibody and an HRP-Avidin conjugate. Visualisation relies on a TMB substrate producing a blue-coloured product that changes to yellow after addition of an acidic stop solution, with absorbance read at 450 nm on a microplate reader. The concentration of Wnt3 in each sample is calculated by reference to a standard curve.

Sensitivity is 0.062 ng/ml with an assay range of 0.156–10 ng/ml. Total assay time is 4 h 30 min, and detection is colourmetric. Kit components are to be stored at +4°C or −20°C as instructed and must not be used past the stated expiration date. The kit is suitable for serum, plasma and tissue homogenates.

Reported average recovery values across the matrix are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Reported linearity upon dilution at 1:2, 1:4, 1:8 and 1:16 is 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5 for each).
