Rat Von Willebrand Factor ELISA Kit

From Antibodies.com, the Rat Von Willebrand Factor ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of rat Von Willebrand Factor in plasma. The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips, with colourimetric detection by reading the absorbance at 450 nm in a microplate reader. The assay range is 15.6-1000 ng/ml and the sensitivity is <6.6 ng/ml, with a total protocol time of 4 hours 30 minutes.

Plasma samples and standards are added to the coated wells where Von Willebrand Factor is captured by the immobilised antibody. After washing, a biotinylated anti-Von Willebrand Factor antibody binds the captured analyte, followed by addition of an HRP-avidin conjugate. TMB substrate is then added to visualise the enzymatic reaction; the reaction is stopped with an acidic solution that produces a yellow colour whose intensity is directly proportional to the amount of analyte present, with concentration calculated from a standard curve. Reported sample recovery values are serum 80%-102% (average 91%, n=5), EDTA plasma 81%-100% (average 90%, n=5) and heparin plasma 80%-89% (average 84%, n=5). Dilution linearity data are also reported for each sample matrix at serial dilutions ranging from 1:2 to 1:16.

Kit components include the pre-coated microplate, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored between +4°C and -20°C as indicated, and the kit should not be used past its stated expiration date.
