Description

Manufactured under the Antibodies.com brand, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is intended for the in vitro quantitative determination of rat Vasoactive Intestinal Peptide (VIP) in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) and is read on a colorimetric microplate reader at 450 nm. Total assay time is 2 hours 30 minutes.

The kit reports a sensitivity of 2.64 pg/ml and an assay range of 7.81-500 pg/ml. The principle employs a competitive enzyme immunoassay technique: VIP present in samples or standards competes with immobilized VIP antigen for binding sites on a Biotinylated Anti-VIP Antibody. After washing, an HRP-Avidin conjugate is added, and the resulting TMB substrate signal — read at 450 nm following the addition of acidic stop solution — is inversely proportional to the concentration of VIP in each sample or standard.

Validation data show average recoveries of 91% for serum (n=5; range 80-102%), 90% for EDTA plasma (n=5; range 81-100%) and 84% for heparin plasma (n=5; range 80-89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries of 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
