Rat Urocortin 2 ELISA Kit

A sandwich ELISA from Antibodies.com enables quantitative in vitro determination of rat Urocortin 2 (UCN2/SRP) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique using an antibody specific for UCN2/SRP pre-coated onto a microplate. Standards and samples are added to the wells, where UCN2/SRP is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-UCN2/SRP Antibody binds the captured analyte, and an HRP-Avidin conjugate is then added. TMB substrate produces a blue colour that converts to yellow upon addition of an acidic stop solution; absorbance is read at 450 nm in a microplate reader and concentration is calculated by reference to the standard curve.

The kit has a stated sensitivity of <5.3 pg/ml and a working range of 15.6–1000 pg/ml, with a total assay time of 4h 30m and colourimetric detection on a pre-coated microplate platform. Kit components include the pre-coated plate, lyophilized standard, two detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4°C or −20°C as instructed.

Recovery has been evaluated across sample types (n=5): serum 80–102% (average 91%), EDTA plasma 81–100% (average 90%), and heparin plasma 80–89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5) was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
