Rat Urocortin 1 ELISA Kit

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Detection is colorimetric, with absorbance read at 450 nm in a microplate reader and the Urocortin concentration calculated by reference to a standard curve. Total assay time is approximately 4 hours 30 minutes.

In the sandwich immunoassay procedure, standards and samples are added to wells coated with an antibody specific for Urocortin. Captured analyte is detected using a Biotinylated Anti-Urocortin Antibody followed by an HRP-Avidin conjugate. TMB substrate is then added; HRP catalysis produces a blue-coloured product that turns yellow after addition of acidic stop solution, with the density of yellow proportional to the amount of Urocortin bound in each well.

Reported performance figures include an assay range of 15.6–1000 pg/ml and a sensitivity of <6.5 pg/ml. Spiked recovery in serum averaged 91% (range 80–102%, n=5), in EDTA plasma 90% (81–100%, n=5), and in heparin plasma 84% (80–89%, n=5). Linearity upon dilution has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma samples.
