Rat Uncoupling Protein 1, Mitochondrial ELISA Kit

This sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat Uncoupling Protein 1 (UCP1), mitochondrial, in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. It is reactive with rat samples and runs on a pre-coated 96-well microplate (12 x 8 well strips) read colorimetrically at 450 nm, with a total assay time of 4 hours 30 minutes.

Standards and test samples are added to the antibody-coated wells; captured UCP1 is detected using a Biotinylated Anti-UCP1 Antibody followed by an HRP-Avidin conjugate and TMB substrate, producing a yellow-coloured product whose absorbance is proportional to the amount of UCP1 in each well. Kit components include a pre-coated 96-well microplate, lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers, stored at +4 °C or −20 °C as instructed.

The assay has a quantitative detection range of 0.312–20 ng/ml and a sensitivity of <0.119 ng/ml. Reported average recovery is 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (range 81–100%, n=5) and 84% in heparin plasma (range 80–89%, n=5), with dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions reported for all three sample types.
