A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com for the in vitro quantitative determination of rat CD40 in serum, plasma or other biological fluids. The kit is supplied as a pre-coated 96-well microplate in a 12 x 8 well strip configuration and is read on a colorimetric microplate reader at 450 nm.
The assay has a stated sensitivity of <5.2 pg/ml and an assay range of 15.6–1000 pg/ml, with a total assay time of 4h 30m. Reported mean recoveries in neat samples are 91% for serum (range 80%–102%, n=5), 91% for EDTA plasma (range 81%–101%, n=5) and 84% for heparin plasma (range 80%–89%, n=5). Dilutional linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma (n=5 per matrix).
Components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date. The protocol uses a biotinylated anti-CD40 detection antibody followed by an HRP–avidin conjugate, with TMB substrate developed using an acidic stop solution to produce a yellow colour whose intensity is proportional to CD40 concentration.
