Rat Tumor Necrosis Factor Receptor Superfamily, Member 1A ELISA Kit

Antibodies.com supplies this sandwich ELISA kit for the in vitro quantitative determination of rat TNFRSF1A in serum, plasma, tissue homogenates, cell lysates and other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, with samples and standards added to the wells before incubation with a biotinylated anti-TNFRSF1A antibody, followed by an HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue colour that turns yellow upon addition of the acidic stop solution, with the density of yellow proportional to the amount of TNFRSF1A captured. Absorbance is read at 450 nm on a colorimetric microplate reader and the concentration calculated from the standard curve.

The stated assay range is 0.156-10 ng/ml with a sensitivity of <0.062 ng/ml, and the protocol runs in approximately 4 hours 30 minutes. Spike-and-recovery testing (n=5) gave averages of 91% in serum (range 83-100%), 94% in EDTA plasma (range 90-98%) and 95% in heparin plasma (range 89-102%). Linearity was demonstrated across serial dilutions, with serum recoveries of 83-110%, 79-100%, 90-98% and 82-108% at 1:2, 1:4, 1:8 and 1:16 respectively; EDTA plasma values of 90-98%, 90-105%, 73-109% and 87-91%; and heparin plasma values of 88-103%, 77-105%, 95-98% and 87-110%.
