From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat TWEAK (Tumor Necrosis Factor Ligand Superfamily, Member 12) in serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay employs a pre-coated microtiter plate with an antibody specific for TWEAK immobilised on the wells. Standards and samples are added, followed by a biotinylated anti-TWEAK detection antibody, HRP-Avidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that converts to yellow upon addition of the acidic stop solution, and the absorbance is read at 450 nm on a colorimetric microplate reader.

The assay provides a quantitative measurement range of 78.1-5000 pg/ml with a reported sensitivity of 25 pg/ml, and the complete protocol takes 4 hours 30 minutes. Recovery has been validated in serum, EDTA plasma and heparin plasma with average values of 91%, 94% and 95% respectively (n=5), and dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across these matrices. Kit components are stored at +4°C or -20°C according to the manual, and the reagents should not be used beyond the stated expiration date.
