The Trypsinogen Activation Peptide (TAP) is the short N-terminal hexapeptide cleaved from trypsinogen during its conversion to active trypsin by enterokinase in the duodenum. When trypsinogen is prematurely activated within pancreatic acinar cells, as occurs in experimental models of acute pancreatitis, TAP is released into the circulation, providing a quantitative readout of pancreatic acinar injury and disease severity.

This Antibodies.com kit is a 96T competitive ELISA developed for rat samples and intended for research use only. The microplate is supplied pre-coated, and the assay relies on competition between TAP present in standards or unknowns and a labelled TAP conjugate for binding to the anti-TAP antibody. After incubation and washing, the chromogenic reaction is read on a standard plate reader, and sample concentrations are interpolated from the calibration curve. A 50 uL sample volume is typical per well.

Typical applications include time-course studies in caerulein- or L-arginine-induced rat pancreatitis models, evaluation of therapeutic candidates intended to limit acinar injury, and characterisation of pancreatic exocrine function in dietary or genetic intervention studies. The kit contains sufficient reagents for one 96T plate. Components should be refrigerated upon arrival and equilibrated to room temperature before use; for longer-term storage, the conjugate and standards may be aliquoted and frozen to limit repeated freeze-thaw cycles.
