Rat Troponin T Type 1, Slow Skeletal ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat Troponin T1/TNT in serum, plasma or other biological fluids.

The assay employs a 96-well microtiter plate pre-coated with an antibody specific for Troponin T1/TNT. Standards and test samples are added to the wells and the target analyte present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Troponin T1/TNT Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution produces a blue colour that turns yellow upon addition of acidic stop solution; the intensity of the yellow colour is proportional to the amount of Troponin T1/TNT captured. The concentration in each sample is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The kit offers a sensitivity of 5.2 pg/ml and an assay range of 15.6-1000 pg/ml, with a total assay time of 4 hours 30 minutes. Reported recovery values are: serum average 91% (range 80%-102%, n=5); EDTA plasma average 90% (range 81%-100%, n=5); heparin plasma average 84% (range 80%-89%, n=5). Dilution linearity for serum (n=5) ranges from 74% to 107% across dilutions of 1:2, 1:4, 1:8 and 1:16; for EDTA plasma (n=5) from 79% to 108%; and for heparin plasma (n=5) from 82% to 105%. The platform is a pre-coated 12 x 8 well strip microplate, with colourimetric detection. Kit components include a pre-coated microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers.
