This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat TRPM7 (Transient Receptor Potential Cation Channel Subfamily M, Member 7) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for TRPM7 pre-coated onto the microplate wells.

Standards and samples are added to the wells, where TRPM7 binds to the immobilized capture antibody. After incubation and washing, a Biotinylated Anti-TRPM7 Antibody is added, followed by incubation with an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The concentration of TRPM7 is calculated by reading the optical density absorbance at 450 nm in a microplate reader and comparing values to the standard curve.

The assay has a sensitivity of 0.052 ng/ml and a product range of 0.156-10 ng/ml. Detection is colorimetric, and the total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
