Rat Transferrin ELISA Kit

Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of rat Transferrin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for Transferrin pre-coated onto a 96-well microplate. Standards and test samples are added into the wells and the Transferrin present is bound by the immobilized antibody. After washing, a Biotinylated Anti-Transferrin Antibody binds the captured Transferrin, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Transferrin captured per well, and concentration is calculated from absorbance at 450 nm against the standard curve.

The pre-coated microplate is arranged as 12 x 8 well strips and read on a colourmetric microplate reader. The assay has a product range of 0.312-20 ng/ml and a sensitivity of <0.143 ng/ml, with a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Reported sample recovery values are: serum (n=5) ranging from 80% to 102% with an average of 91%, EDTA plasma (n=5) from 81% to 100% averaging 90%, and heparin plasma (n=5) from 80% to 89% averaging 84%. Dilution linearity is provided at 1:2, 1:4, 1:8 and 1:16 for serum (87-91%, 87-107%, 74-101%, 92-97%), EDTA plasma (90-105%, 84-101%, 90-101%, 79-108%) and heparin plasma (84-95%, 92-105%, 82-105%, 89-91%) sample types.
