Rat Tissue Inhibitors Of Metalloproteinase 1 ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of rat Tissue Inhibitor of Metalloproteinase 1 (TIMP1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate onto which an antibody specific for TIMP1 has been immobilised; standards and test samples are added to the wells and any TIMP1 present is captured by this antibody. After washing, a biotinylated anti-TIMP1 detection antibody is added, followed by an HRP-Avidin conjugate, and bound enzyme is visualised with TMB substrate. The colour intensity is read at 450 nm on a colorimetric microplate reader and the TIMP1 concentration is calculated by reference to the standard curve.

The kit has a reported product range of 0.156–10 ng/ml and a sensitivity of less than 0.055 ng/ml, with a total assay time of 4 hours 30 minutes. Across rat sample matrices (n=5), average values were 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Parallel dilution data at 1:2, 1:4, 1:8 and 1:16 dilutions gave ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components are stored at +4 °C or –20 °C as indicated, and the reagents must not be used past the stated expiration date.
