From Antibodies.com, this sandwich-format ELISA kit enables the in vitro quantitative determination of rat Tissue Factor Pathway Inhibitor (TFPI) in plasma, urine, tissue homogenates, cell culture supernates or other biological fluids. The assay is performed on a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are captured by an immobilised anti-TFPI antibody, then detected with a Biotinylated Anti-TFPI Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution; absorbance is read at 450 nm on a colorimetric microplate reader.

The reported assay sensitivity is 0.66 ng/ml and the standard curve range is 1.56-100 ng/ml. Total assay time is 4 hours 30 minutes. Recovery was evaluated across serum (n=5, average 91%), EDTA plasma (n=5, average 90%) and heparin plasma (n=5, average 84%), with individual ranges of 80%-102%, 81%-100% and 80%-89% respectively. Linearity of dilution was assessed at 1:2, 1:4, 1:8 and 1:16 ratios: serum (n=5) returned 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma (n=5) returned 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma (n=5) returned 84-95%, 92-105%, 82-105% and 89-91%.

Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date. TFPI is also known by the synonyms EPI, Extrinsic pathway inhibitor, LACI, Lipoprotein-associated coagulation inhibitor and TFPI1.
