A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat Thyroid Hormone Receptor alpha in tissue homogenates or other biological fluids. Quantification is performed on a colorimetric microplate reader, with absorbance read at 450 nm against a standard curve. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

The assay is built around a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells and the captured Thyroid Hormone Receptor alpha is detected using a biotinylated anti-Thyroid Hormone Receptor alpha antibody, followed by an HRP-avidin conjugate and TMB substrate. The reaction is stopped with an acidic stop solution, producing a yellow colour whose intensity is proportional to the amount of analyte bound in each well.

Reported performance figures include a sensitivity of <0.268 ng/ml, an assay range of 0.625–40 ng/ml and a total assay time of 4h 30m. Average recovery values for spiked samples are 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (range 81–100%, n=5) and 84% in heparin plasma (range 80–89%, n=5). Dilutional linearity data covering 1:2 to 1:16 dilutions across serum, EDTA plasma and heparin plasma is provided.
