Rat Thrombospondin 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Thrombospondin 1 in plasma. The kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with absorbance read at 450 nm on a colorimetric microplate reader. Reported sensitivity is <5.3 pg/ml, and the assay range spans 15.6–1000 pg/ml. Total assay time is 4 h 30 min. Kit components are stored at +4 °C or –20 °C as directed in the protocol manual, and the kit should not be used past its expiration date.

Validated spike-and-recovery data are provided for serum (n=5) ranging 80%–102% with an average of 91%; EDTA plasma (n=5) ranging 81%–100% with an average of 90%; and heparin plasma (n=5) ranging 80%–89% with an average of 84%. Linearity-of-dilution values across 1:2, 1:4, 1:8 and 1:16 dilutions are also reported for serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%), each at n=5. The protocol uses a biotinylated anti-Thrombospondin 1 detection antibody and an HRP-avidin conjugate, with TMB substrate producing a blue product that turns yellow on addition of acidic stop solution; the intensity of yellow is proportional to Thrombospondin 1 concentration and is calculated by reference to a standard curve.
