Rat Thrombopoietin ELISA Kit

From Antibodies.com, this sandwich ELISA enables the in vitro quantitative determination of rat Thrombopoietin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with a Thrombopoietin-specific capture antibody. Standards and samples are added, followed by a biotinylated anti-Thrombopoietin detection antibody, an HRP-avidin conjugate and TMB substrate, and the absorbance is read at 450 nm in a colorimetric microplate reader. The Thrombopoietin concentration in each well is then calculated from a standard curve.

The standard curve covers a product range of 15.6–1000 pg/ml with a sensitivity of <5.3 pg/ml, and the total assay time is 4 h 30 min. Across five replicates, average recovery values were 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Linearity of dilution has been validated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components should be stored at +4 °C or −20 °C as instructed, and the kit must not be used past its expiration date.
