Description

From Antibodies.com, this Rat Prosurfactant Protein C ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of rat Prosurfactant Protein C in tissue homogenates, cell lysates, lung lavage fluid or other biological fluids. The assay is reactive with rat and is read colourmetrically on a standard microplate reader.

The assay delivers a sensitivity of 0.147 ng/ml across a working range of 0.312–20 ng/ml, with a total run time of approximately 4h 30m. The kit is built around a pre-coated 96-well microplate arranged as 12 x 8 well strips and uses a biotinylated anti-Prosurfactant Protein C detection antibody followed by an HRP-avidin conjugate, visualised with TMB substrate that yields a yellow product after the addition of stop solution. Standards and samples are added to coated wells, washed, incubated with the biotinylated antibody and then with HRP-avidin, and the concentration of Prosurfactant Protein C in each sample is calculated from the O.D. absorbance at 450 nm against the standard curve. Kit components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

Reported sample recovery is 80–102% (average 91%) in serum, 81–100% (average 90%) in EDTA plasma, and 80–89% (average 84%) in heparin plasma (n=5 for each). Linearity on dilution has been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions, with reported ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
