From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat Surfactant Protein A/PSAP in serum, plasma, lung lavage fluid, tissue homogenates or other biological fluids [C0, S1].

The kit employs a pre-coated microplate (12 x 8 well strips) read at 450 nm on a colorimetric microplate reader. According to the manufacturer's product range, the assay spans 15.6–1000 pg/ml with a sensitivity of <6.9 pg/ml, and total assay time is 4 hours 30 minutes. Recovery in serum ranges 80%–102% (n=5, average 91%); EDTA plasma 81%–100% (n=5, average 90%); heparin plasma 80%–89% (n=5, average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 yields 87–91%, 87–107%, 74–101% and 92–97% in serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% in EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% in heparin plasma (n=5) [S1].

The assay uses a biotinylated anti-Surfactant Protein A/PSAP detection antibody and HRP-avidin with TMB substrate; the resulting yellow colour produced after addition of acidic stop solution is proportional to analyte concentration in each well.
