Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of rat Stromal Cell Derived Factor 1 (SDF1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for SDF1 pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, where any SDF1 present is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-SDF1 Antibody binds the captured SDF1, and an HRP-Avidin conjugate is then added. After further incubation and washing steps, TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of the acidic stop solution. The density of the yellow colour is proportional to the amount of SDF1 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The stated product range is 0.312–20 ng/ml with a sensitivity of 0.125 ng/ml, read on a colorimetric microplate reader. Reported recovery in spiked samples includes serum (range 80–102%, average 91%, n = 5), EDTA plasma (range 81–100%, average 90%, n = 5) and heparin plasma (range 80–89%, average 84%, n = 5). Dilution linearity has been characterised at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma (n = 5), and the total assay time is approximately 4 h 30 min.

Kit components comprise a pre-coated 96-well microplate (12 × 8 well strips), lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30×), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Kit components should be stored at +4 °C or −20 °C as instructed, and the kit must not be used past its expiration date.
