Rat Spondin 2 ELISA Kit

Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of rat SPON2 in serum, plasma or other biological fluids. The kit reacts with rat samples and uses a colorimetric detection format read on a microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

The assay uses a microplate pre-coated with an antibody specific for SPON2. Standards and test samples are added to the wells; following incubation and washing, a biotinylated anti-SPON2 antibody is applied, followed by an HRP-avidin conjugate. TMB substrate is then added; catalysis produces a blue product that turns yellow upon addition of the acidic stop solution, with the density of yellow proportional to the amount of SPON2 captured. The concentration of SPON2 is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Performance specifications include a product range of 0.156–10 ng/ml and a sensitivity of <0.062 ng/ml, with a total assay time of 4h 30m. In spike-and-recovery testing (n=5), averages ranged from 84% to 91% across serum (80–102%), EDTA plasma (81–100%) and heparin plasma (80–89%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions, with serum recoveries of 87–91%, 87–107%, 74–101% and 92–97%; EDTA plasma at 90–105%, 84–101%, 90–101% and 79–108%; and heparin plasma at 84–95%, 92–105%, 82–105% and 89–91% (n=5 per sample type).
