Rat Spleen Focus Forming Virus Proviral Integration Oncogene ELISA Kit

This Sandwich ELISA kit from Antibodies.com is intended for the in vitro quantitative determination of rat PU.1/Spi1 — also referred to as the Spleen Focus Forming Virus Proviral Integration Oncogene — in tissue homogenates, cell lysates and other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, an immobilised antibody specific for the target, a biotinylated anti-PU.1/Spi1 detection antibody, and an HRP-avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that changes to yellow on addition of acidic stop solution; absorbance is read at 450 nm on a colorimetric microplate reader.

The kit has a stated sensitivity of <0.116 ng/ml and a working range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Reported average recovery values are 91% in serum (n=5, range 80%–102%), 90% in EDTA plasma (n=5, range 81%–100%) and 84% in heparin plasma (n=5, range 80%–89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is also reported for these sample types: serum 87–91%, 87–107%, 74–101%, 92–97%; EDTA plasma 90–105%, 84–101%, 90–101%, 79–108%; heparin plasma 84–95%, 92–105%, 82–105%, 89–91% (all n=5). Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or -20°C as instructed.
