Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat SGPP1 (Sphingosine-1-Phosphate Phosphatase 1) in tissue homogenates or other biological fluids.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for SGPP1 is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where SGPP1 is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-SGPP1 antibody binds the captured target, after which an HRP-avidin conjugate is added. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and SGPP1 concentration is calculated by reference to a standard curve.

Reported sensitivity is <0.057 ng/ml with an assay range of 0.156-10 ng/ml. The assay is reactive with rat samples and is suitable for tissue homogenates, with a total assay time of 4 hours 30 minutes. Recovery data for serum (average 91%, range 80%-102%), EDTA plasma (average 90%, range 81%-100%), and heparin plasma (average 84%, range 80%-89%) are provided across n=5 samples. Dilution linearity data are also supplied for 1:2, 1:4, 1:8, and 1:16 dilutions in each sample matrix. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
