Description

From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of rat Somatostatin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with Somatostatin antigen and is reactive with rat samples.

The principle is a competitive immunoassay: Somatostatin present in samples or standards competes with the immobilized antigen for binding sites on a Biotinylated Anti-Somatostatin Antibody. After washing to remove unbound antibody, an HRP-Avidin conjugate is added and the plate is incubated. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader, and the yellow intensity is inversely proportional to the amount of Somatostatin present, which is calculated by reference to a standard curve.

Performance specifications include a quantitative range of 7.81-500 pg/ml and a sensitivity of 2.16 pg/ml, with a total assay time of 2 hours 30 minutes. Sample recovery has been validated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5) and heparin plasma (range 80%-89%, average 84%, n=5). The kit is supplied with pre-coated microplate strips, lyophilized standard, detection reagents, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with components stored at +4°C or −20°C as instructed.
