Rat Sirtuin 3 ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of rat Sirtuin 3 (SIRT3) in tissue homogenates, cell lysates, or other biological fluids. The microtitre plate is pre-coated with an antibody specific for SIRT3. Standards and test samples are added to the wells, where SIRT3 is captured by the immobilised antibody. After washing, a Biotinylated Anti-SIRT3 Antibody binds the captured SIRT3, followed by incubation with an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of SIRT3 captured in each well. The concentration is calculated from the O.D. absorbance at 450 nm read in a colorimetric microplate reader, with reference to a standard curve.

The kit has a sensitivity of <0.088 ng/ml and a measuring range of 0.312–20 ng/ml. The total assay time is 4 hours 30 minutes. Kit components should be stored at +4 °C or –20 °C as instructed and not used past the expiration date. Validation data report average recoveries of 91% for serum, 90% for EDTA plasma, and 84% for heparin plasma across the tested sample set (n=5 per type).
