The kit employs the sandwich enzyme immunoassay technique: an antibody specific for NEU2 is pre-coated onto a microtiter plate, standards and test samples are added, and the captured target is detected using a Biotinylated Anti-NEU2 Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450nm in a microplate reader.

Reported performance characteristics include a sensitivity of 0.059 ng/ml and a measuring range of 0.156–10 ng/ml. The colourimetric assay has a total run time of 4h 30m. Sample recovery has been evaluated across matrices, with averages of 91% for serum, 90% for EDTA plasma, and 84% for heparin plasma, and dilution linearity reported from 1:2 to 1:16 across the same sample types.
