Rat Semaphorin 3A ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat Semaphorin 3A in serum, plasma, tissue homogenates, and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and follows the sandwich immunoassay principle: Semaphorin 3A in standards and samples is captured by an immobilised antibody, detected with a biotinylated anti-Semaphorin 3A antibody, and visualised via an HRP-Avidin conjugate with TMB substrate. Absorbance is read at 450 nm on a colorimetric microplate reader, and the quantitative assay has a product range of 0.156–10 ng/ml with a sensitivity of <0.057 ng/ml. The total assay time is 4 hours 30 minutes.

Recovery was measured in serum (range 83–100%, average 91%), EDTA plasma (range 91–99%, average 95%) and heparin plasma (range 90–99%, average 94%), with n=5 for each matrix. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across these sample types. The kit is supplied with a pre-coated microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers; components are stored at +4°C or -20°C as indicated.
